Boozer is out with the strained left quadriceps tendon he suffered in Wednesday's victory over Milwaukee. Williams is doubtful with a sprained left ankle, although he did take part in the pregame shootaround.
In addition to Williams and Boozer, the Jazz also will play without Kyle Korver, who aggravated a right wrist injury when he fell in Wednesday's game, and Jarron Collins, who continues to nurse an injured right elbow.
That would leave the Jazz with only 11 players in uniform, though the Spurs are expected to play without Manu Ginobili and Tony Parker.
Jazz coach Jerry Sloan said he would start rookie forward Kosta Koufos in Boozer's place.
"I've known for years we've been really lucky," Sloan said. "We haven't had a lot of injuries. If you have injuries, what do you do about it. it's a crybaby thing as far as I'm concerned. You've just got to go play. That's what everybody else does."
As well as Paul Millsap has played, Sloan is reluctant to change his role coming off the bench. Same thing for Andrei Kirilenko. Koufos will match up against Fabricio Oberto while Mehmet Okur takes Tim Duncan.
"We have to have somebody to play in that second group," Sloan said.
"Those guys have done a great job playing off the bench for us. When they come off the bench, they've been pretty effective."
Brevin Knight is expected to return after missing Wednesday's game with a strained left groin. That gives the Jazz two point guards in Knight and Ronnie Price; Price was the only available point guard for Wednesday's game.
Williams had missed only four games in three previous seasons due to injury. Boozer coincidentally was out for three of those games - - two in the 2005-06 season and one in February 2007.
